INFORMATION ABOUT THIS DATA

Data Set Name 1. Virulence and Persistence Data for Salmonella Typhimurium using the human surrogate host C. elegans
Data Set Description Virulence and persistence studies using RWV-cultured S. Typhimurium and the human surrogate host, Caenorhabditis elegans.

Virulence Experiment 1. These are the raw and processed data for the first biological replicate (of two) of a virulence study performed with wild type C. elegans as the host (N2 Bristol strain) and the foodborne pathogen, wild type S. Typhimurium (strain ??3339) that had been cultured in the Rotating Wall Vessel (RWV) bioreactor to late log/early stationary phase prior to infection. E. coli strain OP50 served as a non-pathogenic control. Samples are labeled with a number (-1, -2, -3), representing technical replicates within the experiment (individual wells). See the "Experimental Detail" tab for additional detail.

Virulence Experiment 2. These are the raw and processed data for the second biological replicate (of two) of a virulence study performed with wild type C. elegans as the host (N2 Bristol strain) and the foodborne pathogen, wild type S.

Virulence Experiment 3. These are the raw and processed data for the first biological replicate (of two) of a virulence study performed with the ?pmk-1 mutant strain of C. elegans as the host and the foodborne pathogen, wild type S.

Virulence Experiment 4. These are the raw and processed data for the second biological replicate (of two) of a virulence study performed with the ?pmk-1 mutant strain of C. elegans as the host and the foodborne pathogen, wild type S.

Measurement or Analysis Technique
S. Typhimurium was cultured to late log/early stationary phase in the Rotating Wall Vessel (RWV) bioreactor positioned in the LSMMG and control (1xg) positions. Nematodes were briefly exposed to the RWV-cultured pathogen or a non-pathogenic control (Escherichia coli OP50) in liquid and then transferred to lawns of OP50. For virulence studies, nematodes were assessed each day for viability and transferred to new lawns accordingly until the end of the study. For persistence studies, nematodes were infected as described above and on the designated day of testing were washed and homogenized to plate for viable colony forming units.
