Virulence and persistence studies using RWV-cultured
B. multivorans and the human surrogate host, Caenorhabditis elegans.
Virulence Experiment 1. These are the raw and processed data for the first biological replicate (of two) of a virulence study performed with wild type C. elegans as the host (N2 Bristol strain) and an ISS potable water isolate, B. multivorans, that had been cultured in the Rotating Wall Vessel (RWV) bioreactor to stationary phase prior to infection. E. coli strain OP50 served as a non-pathogenic control. Samples are labeled with a number (-1, -2, -3), representing technical replicates within the experiment (individual wells). See the "Experimental Detail" tab for additional detail. Note: two additional OP50 technical replicates are not shown due to their exclusion from the trial due to potential contamination. B. multivorans (a potable water isolate from the International Space Station) was cultured to stationary phase in the Rotating Wall Vessel (RWV) bioreactor positioned in the LSMMG and control (1xg) positions. Nematodes were briefly exposed to the RWV-cultured pathogen or a non-pathogenic control (Escherichia coli OP50) in liquid and then transferred to lawns of OP50. For virulence studies, nematodes were assessed each day for viability and transferred to new lawns accordingly until the end of the study. For persistence studies, nematodes were infected as described above and on the designated day of testing were washed, homogenized and plated for viable colony forming units.
